The nuclear envelope (NE) is composed of the outer nuclear membrane (ONM) and the inner nuclear membrane (INM), which are separated from the perinuclear space and joined with nuclear pore complexes.
INM proteins play crucial roles in cell cycle regulation, DNA repair, aging and cell migration of tumor formation and progression. However, the specific INM proteins and INM protein-based mechanisms that contribute to cancer aggression have yet to be studied.
The results of the Research have been published in the scientific journal Oncogene.
INM 201 protein: why this is an important discovery
In addition to the classic functions of providing physical strength and stability to protect the core, INM proteins link to cancer pathogenesis.
In this study, the researchers found that INM 201 TMEM201, an INM type III protein, was highly expressed and predicted poor breast cancer survival in invasive breast cancer.
Scientists then investigated the increased expression of INM 201 (TMEM201) which promoted TGFβ-mediated epithelium-mesenchymal transition (EMT) by facilitating BC cell migration, invasion and survival properties. The study showed that TMEM201 physically interacted with SMAD2 / 3 and was required for SMAD2 / 3 phosphorylation.
These results revealed the role of the specific INM 201 protein in aggressive BC metastasis and suggested a potential therapeutic strategy for poor-prognosis disease.
This study not only reveals that the INM 201 protein acts as a positive regulator of signaling TGFβ interacting with SMAD2 / 3 in breast cancer metastases, but also broadens understanding of the involvement of the INM protein in cancer cell biology and oncology as a whole.
Breast cancer is the most common cancer in women and the second most common cancer overall. There were over 2 million new cases in 2018.
Breast cancer develops due to genetic mutations or damage to the DNA. These can be exposure to estrogen, hereditary genetic defects, or hereditary genes that can cause cancer, such as genes BRCA1 And BRCA2 .
When a person is healthy, their immune system attacks any abnormal DNA or growths. When a person has cancer, this doesn’t happen.
As a result, the cells within the breast tissue begin to multiply uncontrollably and do not die as usual. This excessive cell growth forms a tumor that deprives surrounding cells of nutrients and energy.
Breast cancer usually begins in the inner lining of the milk ducts or in the lobules that supply them with milk. From there, it can spread to other parts of the body.
This year (2021), approximately 284,200 people (281,550 women and 2,650 men) in the United States will be diagnosed with invasive breast cancer. Breast cancer is the most common cancer in women, excluding skin cancer.
6% of women have metastatic breast cancer when it is first diagnosed. More research is needed to determine how many people with non-metastatic breast cancer subsequently develop metastatic breast cancer.
Breast cancer is the second most common cause of cancer death in women. It is estimated that 44,130 people (43,600 women and 530 men) will die from breast cancer this year. Metastatic breast cancer will cause the vast majority of these deaths.
The 5-year survival rate tells you what percentage of people live at least 5 years after cancer is discovered. Percent means how many in 100. The 5-year survival rate for women with metastatic breast cancer is 28%. The 5-year survival rate for men with metastatic breast cancer is 22%.
It is important to remember that breast cancer is treatable at any stage. Treatments for metastatic breast cancer are continuously improving and have been shown to help people with metastatic breast cancer live longer with a better quality of life.
Furthermore, statistics on survival rates for people with metastatic breast cancer are an estimate. The estimate comes from annual data based on the number of people with this cancer in the United States. Additionally, experts measure survival statistics every 5 years.
Therefore the estimate may not show the results of a better diagnosis or treatment available for less than 5 years. Talk to your doctor if there are any questions about this information.
